Technical principle:
We have developed a very simple process. Using our glass capillary-based PDC technology, a small amount of micron cell suspension can be sucked with almost no dead volume. Then, use patented dispensing/imaging technology to monitor the dispensing of each drop of liquid medicine. Our PDC dispensing technology usually dispenses a droplet volume of 100-300 picoliters. Since each droplet will be ejected, we monitor it for the presence of a single unit. If a cell exists, we store it in the target vessel of your choice. It may be a micro unit, a micron plate, a slide or other things you like. If there are no cells in the droplet, or there are multiple cells, we will recycle the droplet to ensure that your precious rare cells are not wasted.

Application case
HuBBBTM technology:
HuBBBTM technology is a very powerful B cell immortalization system, but only 10% of immortalized B cells are successfully cloned, so the data is still very limited. The combination of HuBBBTM and CELLEN ONE significantly increases the cloning efficiency to 90%, saving a lot of time and ensuring the identification of the best clones.
Cloning field:
Purpose: To demonstrate the ability to clone cells for mAb development (e.g. CHO, hybridoma)
Difficulty: Ensure the monoclonality and viability of cells isolated and cultured individually
CellenONE method: Print single cells on a microscope slide, verify monoclonality, clone recovery/survival by distributing single clones into 96-well plates, and counting n colonies/survival after 7 days.
FACS vs CellenONEX1:
1. Comparison purpose: compare the clone recovery of BDfacsaria 3 and CellenONEX1
2. Experimental cells: CHO cell transformation with GFP-negative reporter gene and drug resistance gene fusion-related genes. During the experiment, 70% of CHOs were GFP negative.
3. Experimental details: The cloning is carried out in a 96-well plate, and each well is supplemented with 200μl medium with adrenaline selective antibiotics. After culturing for 5 days, the number of wells with at least 5 cells in a single colony was positive. (GFP+ clones were eliminated due to selective existence)
4. Expected result:
i. The success rate after FACS classification is 100% (excluding GFP +ve)
ii. The success rate of CellenONE sorting is 70%.
